Application of PCR-RFLP for Species Identification of Symbiont Insects of Honey Bees

Abstract

Monitoring the species composition of arthropod symbionts in honeybee hives is important and necessary for the health of bee colonies. However, it is difficult to use morphological methods when working with degraded material, body fragments, or preimaginal stages. In this study, a PCR-RFLP method was developed and validated for the primary molecular identification of several species of insect symbionts and pests of honeybees (Vespula germanica, V. vulgaris, Galleria mellonella, Tribolium madens, Forficula tomis) and their differentiation from the honeybee (Apis mellifera). Based on bioinformatics analysis of the consensus sequences of the COI gene, in silico restriction was performed with more than 50 restriction endonucleases, which resulted in the selection of only two (AclWI and BstX2I). Unique, non-overlapping restriction profiles were obtained for each of the six species. Sanger sequencing was performed to validate the developed method; the obtained restriction profiles for each sample coincided with the sequencing results. The method has proven effective for identifying insects at different stages of development and may be a rapid and cost-effective alternative to sequencing for routine monitoring of bee symbionts. This could potentially be useful for developing specific methods for controlling the population of bee hive pests.
