Getting rice regenerantes anther culture in vitro single-stage method

Comparative investigation of two-step and one-step methods of regeneration on anther culture in vitro of Oryza sativa L. subspeciesjaponica Kato variety Lugovoy were carried out. Anthers inoculated on two types of basal mediums N6, contained 2,0 mg / l 2,4- dichlorphenoxyacetic acid (N6-2,4-D - two-step method as control) and 10,0 mg / lphenoxyacetic acid 10,0 mg / l + 0,5 mg / l naphtalenacetic acid (N6-PAA - one-step method). In control, calluses transplanted on basal medium for regeneration. Callusability was higher in three times on medium N6-2,4-D (12,2 %), then on medium N6-ФУК (4,0 %). Indexes of regeneration were much more in control too: after medium N6-2,4-D number of green regenerants per callus - 7,93 and number of productive regenerants per callus - 3,50; on medium N6-PAA number of green regenerants per callus - 0,89 and number of productive regenerants per callus 0,34. Portion of productive regenerants numbered 44% and 39% accordingly. One-step method of regeneration on rice anther culture in vitro is effective. But two-step method suits for Far Eastern genotypes better because of green regeneration and productive regeneration are high. Productive regenerants can utilize in rice breeding program in Russian Far East.Downloads
